Detection Principle:
This kit uses double antibody sandwich enzyme-linked immunosorbent assay technology. The specific anti mouse il-10antibody was pre coated on a high affinity microplate. The standard, sample to be tested and biotinylated detection antibody are added into the wells of the enzyme plate. After incubation, the il-10present in the sample combines with the solid-phase antibody and detection antibody to form an immune complex. After washing to remove unbound material, horseradish peroxidase labeled streptavidin (streptavidin HRP) was added. After washing, the chromogenic substrate was added to avoid light for color development. Stop the reaction by adding stop solution, and determine the absorbance value at 450 nmwavelength (reference correction wavelength 540nmor 570nm)

Background:
Mouse interleukin -10 (IL-10) also known as cytokine synthesis inhibitor (CSIF). It is IL-10 & Alpha-a member of the helical cytokine family, which also includes il-19, il-12, il-22, IL-24 , and il-26/ak155. Many activated hematopoietic stem cells, hepatic stellate cells, keratinocytes, and placental cytotrophoblasts secrete il-10. Human IL-10 is active in mouse cells, but mouse IL-10 does not act on human cells. The amino acid homology of mature mouse IL-10 and rat IL-10 is 85%, and that of cattle, dogs, horses, cats, humans, sheep, and pigs is 70~77%. Mature mouse IL-10 is a noncovalently bound homodimer with a molecular weight of 36 kDa containing two intrachain disulfide bonds.
