Detection Principle:
This kit uses double antibody sandwich enzyme-linked immunosorbent assay technology. The specific anti mouse cxcl2/mip-2 antibody was pre coated on a high affinity microplate. The standard, sample to be tested and biotinylated detection antibody are added into the wells of the enzyme plate. After incubation, cxcl2/mip-2 in the sample is combined with the solid-phase antibody and detection antibody to form an immune complex. After washing to remove unbound material, horseradish peroxidase labeled streptavidin HRP was added. After washing, the chromogenic substrate was added to avoid light for color development. Stop the reaction by adding stop solution, and determine the absorbance value at 450 nm wavelength (refer to the correction wavelength of 540nm or 570nm).

Background:
Chemokine ligand 2 (cxcl2) is a small cytokine belonging to the CXC chemokine family, also known as macrophage inflammatory protein 2-& alpha; (mip2 alpha), growth regulatory protein & amp; beta; (GRO beta) and gro oncogene - 2 (gro-2), cxcl2 / gro beta, also known as MIP-2 in mice and cinc-3 in rats, is a member of the CXC chemokine family. Cxcl2 has 90% homology with CXCL1 in amino acid sequence. This chemokine is secreted by monocytes and macrophages and has a chemotactic effect on polymorphonuclear leukocytes and hematopoietic stem cells. The gene for cxcl2 is located on human chromosome 4 in other CXC chemokine clusters. Cxcl2 mobilizes cells by interacting with a cell surface chemokine receptor called CXCR2.
The length of human cxcl2 / gro beta is 107 amino acids (AA) and the estimated molecular weight is 11 kDa. The mouse and rat orthologs share 70% and 71% amino acid sequence identity with the human protein, respectively. The N-terminal amino acids 1-4 of cxcl2 / gro beta can be post translationally cleaved, thereby enhancing hematopoietic biological activity. Cxcl2 / gro beta is produced by a variety of cell types, including monocytes and macrophages at the site of inflammation, and has chemotactic effects on granulocytes, including neutrophils.
