Dialyzed fetal bovine serum plays a role in these research areas

The dialysis method removes small molecules with molecular weights less than 10 kDa to avoid interference with cells by exogenous small molecules. Small molecules below 10kDa include chemical small molecules, nucleotides, certain hormones, salts, low molecular weight proteins, amino acids, cytokines, glucose, residual antibiotics and other foreign molecules. It is suitable for applications such as pulsed tracing experiments, such as isotope labeling of specific small molecules and addition to culture systems to study cellular metabolism, as well as post-transfection screening applications. It is a great tool for studying proteomics, isotope labeling, cell signaling and reporter gene analysis, and reporter cell line screening.
1. Small molecule labeling experiments
The experiment is to incorporate small molecules such as nucleotides labeled with isotopes into the culture medium to label the newly synthesized DNA in the cell, generally culture for a period of time, the DNA in the cells will be isotopically labeled, and then DNA extraction, purification and other experimental analysis. In order to avoid the interference of natural nucleotides in fetal bovine serum, dialysis fetal bovine serum is required to ensure that the nucleotides incorporated into the cellular DNA are labeled nucleotides.

2. Screening of positive transfected cell clones
Some genetically defective cell lines, such as CHO-DFGRˉ, lack dihydrofolate reductase (DHFR) and must survive in cultures supplemented with hypoxanthine, thymine, and glycine. The gene of interest and the DHFR gene we want to express can be constructed as a plasmid, then transfected into cells and cultured in medium without the above additives. Defective cells that are not transshipped into the recombinant plasmid will undergo apoptosis; Cells that are successfully transferred into the recombinant plasmid survive and form cell clones. In order to avoid the interference of hypoxanthine, thymine and glycine in fetal bovine serum, dialysis fetal bovine serum needs to be used to ensure that the complete culture medium does not contain hypoxanthine, thymine and glycine.

3. Cell platform drug screening
Cell-level drug screening is a drug screening model that is closer to physiological conditions, in which the model is the target cells to be designed for drug action, the cells are obtained by cell culture technology, these cells interact with candidate compounds, and the compounds are screened by measuring the ability of the compounds through detection techniques similar to biochemical level screening. In the process of drug screening, dialysis fetal bovine serum is required to avoid the interference of exogenous small molecules in the experiment.
