      Struggling to Reproduce the Myasthenia Gravis Model? A Guide to AChRα(97–116) Peptide Experimental Applications

      I. What Is the AChRα(97–116) Peptide?

      AChRα(97–116) is a synthetically produced peptide corresponding to residues 97–116 of the rat acetylcholine receptor α subunit. Its amino acid sequence is DGDFAIVKFTKVLLDYTGHI, with a molecular weight of 2252.56 and purity ≥98%, supplied as a white powder. It is a well-recognized immunodominant T-cell epitope peptide. Unlike the full-length AChR protein, this fragment specifically activates T and B lymphocytes and breaks self-immune tolerance; it offers advantages of simple structure, easy purification, and high modeling stability.

      II. What Is the Core Role of AChRα(97–116)?

      The primary application of this peptide is to induce the experimental autoimmune myasthenia gravis (EAMG) animal model, mimicking the pathogenesis of human myasthenia gravis. After immunization, the host produces anti-AChR autoantibodies that disrupt neuromuscular-junction transmission, yielding the characteristic pathological features of muscle weakness and decremental responses upon repetitive stimulation. Compared with crude naturally extracted AChR protein, the Absin peptide abs837315 shows small inter-batch variation, low impurity content, and controllable immunization dose, enabling stable induction of disease in Lewis rats and mice. It is suitable for research directions including autoimmune pathogenesis, pharmacological intervention, and immune tolerance.

      III. Which Experimental Scenarios Use AChRα(97–116)?

      • Animal modeling: Subcutaneous emulsion injection in Lewis rats combined with CFA adjuvant to construct the EAMG model, for myasthenia scoring and repetitive nerve-stimulation testing;
      • In vitro immune-cell experiments: Lymphocyte proliferation assays, T-cell epitope identification, and flow-cytometric immunophenotyping;
      • Molecular immune detection: ELISA for serum anti-AChR antibodies and peptide-specific immuno-blotting;
      • Immune-tolerance intervention: Intranasal peptide instillation to induce mucosal tolerance, investigating autoimmune-suppression strategies.

      This peptide, with its stable purity, is compatible with both in vivo modeling and multi-level in vitro cellular and molecular immune assays.

      Application overview

      The AChRα(97–116) peptide is a core tool for autoimmune-myasthenia research. With its well-defined immune epitope, high purity, and strong modeling reproducibility, it covers the full chain of in vivo animal models, in vitro cells, and molecular immune detection, providing standardized antigen material for research on myasthenia gravis pathogenesis, new-drug screening, and immune-regulation studies.
