Its pH value is stabilized at 7.0-7.6 with osmolality of 320-360mOsm/kg, building the most suitable living environment for cells.

04 Practical Operation Guide for Tissue Preservation Solution
Sample Pre-Treatment
1. Ensure fresh tissues are free of fungal, bacterial and mycoplasma contamination, which is the critical first step;
2. Select viable tissues in appropriate areas, remove necrotic redundant tissues and residual blood;
3. For parenchymal organs, trim tissues into blocks of about 0.5cm width × 0.5cm thickness with surgical scissors or scalpels. Excessively large tissue blocks will hinder the exchange of metabolic waste and nutrients in preservation solution.
Preparation and Usage of Preservation Solution
1. Pre-cool the preservation solution to 2-8°C before use, and keep specimens within this temperature range during transportation and preservation;
2. Calculate the specimen volume, and completely immerse tissues in at least 5 times the volume of tissue preservation solution. For long-term preservation (24-72 hours), 5-10 times volume of solution is recommended.
Post-Preservation Treatment
1. Preserved tissues can be directly applied to follow-up experiments;
2. Some products contain antioxidants. If you need to avoid their experimental interference, rinse tissue samples with normal saline or phosphate buffered saline for 2-3 times.
05 Special Considerations for Different Specimens
- Parenchymal Organs (Liver, Kidney, etc.): Cut into proper sizes (thickness ≤ 0.5cm) to guarantee full penetration of preservation solution and prevent necrosis of central areas caused by insufficient infiltration.
- Cavity Organs (Intestines, Blood Vessels, etc.): Besides external immersion, flush and perfuse the lumen with preservation solution to keep consistent internal and external environment.
- Special Notes:
- Certain tissue preservation solutions perform poorly for blood cell storage, not recommended for blood cell preservation;
- Designed only for tissue storage, not suitable for single-cell preservation; dedicated cell preservation products should be selected for single cells.
06 Common Problems and Solutions
Precipitation Issue
Flocculent precipitation may appear after thawing certain products. Restore the solution to room temperature to fully dissolve precipitates before aliquoting or usage.
Color Change
The solution may turn dark yellowish-brown after long-term storage at 4°C, which normally does not affect performance.
Limitation for Long-Term Storage
Special cryopreservation medium is required for ultra-long storage (ensuring high cell viability after recovery). Tissue preservation solution is designed for short-term low-temperature storage instead of long-term cryopreservation.
Query about Tissue Size
Size requirements vary among different products, some have no strict restrictions. If trimming is necessary, operate inside preservation solution to better protect cells compared with exposure outside the solution.
