Validation data (cell sorting renderings)

 

1. Mouse CD8 + T cell sorting kit (negative selection method) (abs50129)

 

 

CD8 + T cells were sorted from C57BL/6 mouse spleen cells, and the cells before and after sorting were labeled with FITC anti-mouse CD8 antibody (clone No. 53-6.7) and analyzed by flow cytometry. The purity of CD8 + T cells before and after sorting was 13.8% and 97.1%, respectively.

 

2. Human CD8 + T cell sorting kit (positive selection method) (abs50129)

 

 

CD8 + T cells were sorted from human PBMC and subjected to flow cytometric analysis after staining with PE-labeled anti-human CD8a antibody (clone number RPA-T8) and APC-labeled anti-human CD3 antibody (clone number OKT3). The purity of CD3+CD8 + T cells before and after sorting was 15.3% and 91.5%, respectively.

 

Validation data (T cell culture renderings)

 

T cell serum-free complete medium: immune cell serum-free medium (abs9772) + 200-500 U/mL IL-2 (abs04045) + appropriate amount of CD3/CD28 magnetic beads (abs160019)

 

 

CD3 + T cells were stimulated for 24 to 48 hours using human CD3/CD28 T cell activation magnetic beads, and the cells were labeled with FITC anti-human CD69 antibody (clone number FN50) and PE anti-human CD25 antibody (clone number BC96) for flow cytometry analysis. The proportion of CD25 + CD69 + T cells in the total cells before and after stimulation was 0.70% and 75.81%, respectively; T cells-Day5-cell proliferation was obvious.

(1) Fast: it takes at least 15 minutes to sort the target cells;

(2) Simple: no separation column is needed, and the target cells can be quickly sorted by using a magnetic separator;

(3) High purity: the purity of sorted cells can reach more than 95%;

(4) High activity: Cells with high activity, unlabeled with antibodies and magnetic beads, and good functions can be obtained.

(1) The protein type is clear: there are no other protein components except human serum albumin and recombinant human insulin;

(2) Many types of cultured cells: suitable for T, CIK, DC-CIK, TIL, CAR-T, CAR-NK and other cell expansion culture;

(3) Contains antibiotics (kanamycin sulfate) and phenol red: effectively prevent bacterial contamination during cell culture and indicate pH change;

(4) It has a wide range of applicable research: for example, it is used for adoptive immunotherapy applications and experimental research on cellular immune response.
