How to Retrieve Gene CDS Sequences Using NCBI
Many researchers need to synthesize mRNA and obtain the CDS sequence of a gene. Today, let’s explore how to find the CDS sequence of a gene.
1. Open the NCBI website: https://www.ncbi.nlm.nih.gov/2. Click the following option to find “Gene”
3. Select “Gene” and enter the gene name in the search box. The following uses “Human TREM2” as an example. Click “Search”
4. The first result on the search page is the one, as shown in the following figure
5. Click to enter the gene details page. You can see multiple transcripts, as shown in the red box in the following figure.
6. Click “reference sequence details” We need to check the literature citations of the transcripts and select the appropriate transcript based on its length.
7. There are two results.
8. Click to check the CDS sequence length. Generally, the longest sequence is selected. In the following figure, “NM_018965.4” is chosen.
9. Click “CDS”, and the brown-marked sequence that pops up is the CDS sequence of the gene.
