mIHC Literature Interpretation:Biology and Diversity of Glomerular Epithelial Cells.
Parietal epithelial cells (PECs) are a layer of flat epithelium lining the Bowman's capsule (glomerular capsule) and are derived from the same cell lineage as podocytes. In the mature kidney, these two types of cells are anatomically close and are considered "progenitor cells" with the potential to differentiate into podocytes, which has attracted significant attention. Extensive research has shown a close association between PECs and the formation of crescentic glomerulonephritis. On April 23, 2023, Professor Xu Anlong's team from Beijing University of Chinese Medicine published their research findings in the authoritative journal Kidney International, titled "Single cell landscape of parietal epithelial cells in healthy and diseased states." This study established, for the first time, a single-cell map of glomerular PECs, enhancing the understanding of their heterogeneity and biology. It also identified podocyte progenitor cells from PEC subpopulations, revealing the molecular mechanisms of PEC activation and proliferation, which play a crucial role in the pathogenesis and progression of crescentic glomerulonephritis.

The research team utilized single-cell transcriptome analysis combined with histopathological staining and other techniques to conduct an in-depth study of PECs. Based on these findings, they further identified the target proteins Mif and Csf1r that can block the formation of glomerular crescents. They then used animal experiments with the targeted inhibitors caffeic acid and Sotuletinib to verify their effects in inhibiting the formation of crescent glomeruli, providing new insights for the clinical treatment of crescentic glomerulonephritis.
